Hepatocellular IL6/gp130/STAT signalling protects the liver in T-cell mediated hepatitis by a massive up-regulation of SAA protein

To determine the molecular transducers of the IL6/gp130 dependend liver protection in the ConA hepatitis, we generated mice with hepatocyte specific deletions for gp130 STAT or RAS signalling to differentiate the gp130 downstream cascades in this model and to identify potential effecter molecules.To verify the phenotype of the new mice strains on functional level, we isolated primary hepatocytes from ΔSTAT and ΔRAS mice, stimulated them with IL6 and analyzed the gp130 dependend STAT activation and the RAS/MAPK signalling.
