The adipokine leptin is secreted to the blood stream by adipocytes and signals body fat mass to leptin receptors in the brain, mainly located in the hypothalamus. The diabetes allele of the leptin receptor (Lepr db) leads to false splicing, loss of signaling function of the receptor and a disruption of the adipostatic feed back loop between the adipocyte and the brain. Mice homozygous for diabetes (db/db) become obese at around 3 to 4 weeks of age and develop hyperglycaemia insulin resistance at 4–8 weeks of age.
