Introduction: Methylation in the enhancer region of the DNA repair protein MGMT correlates with the outcome of patients treated by alkylating chemotherapeutics. Therefore epigenetic silencing of MGMT inducing decreased expression, avoids repair of the DNA lesions introduced by chemotherapy. For this mechanism a good correlation of protein content and outcome is expected as well, but has not yet been shown in a conclusive fashion.
